ABSTRACT

The objective of this study was to analyze histological and biochemical biomarkers in U. cordatus gills indicative of impacts in São Marcos Bay. Crabs were collected from four areas in São Marcos Bay: A1=Ilha dos Caranguejos (with low impact); A2=Coqueiro, A3=Porto Grande, A4=Cajueiro (potentially impacted areas). The biometric data of each specimen was measured. Gill samples were submitted to standard histological technique and homogenized in phosphate buffer, and the supernatant was used for analysis of glutathione S-transferase (GST) and catalase (CAT) enzyme activity. Biometric data indicated that crabs found in A1 are significantly (P<0.05) larger and heavier than crabs found in A2, A3 and A4 areas. Gill alterations (rupture of pilaster cells, Dilation of the marginal channel, Cuticle Rupture and necrosis) were significantly (P˂0.05) more frequent in the crabs in A2, A3 and A4 than crabs in A1. The enzymatic activities of GST and CAT showed significant difference (P<0.05) between the sampling areas, similar to that observed for gill alterations. The biomarkers analyzed showed that the crabs are under different impact levels (A4> A3> A2> A1) along the São Marcos Bay.(AU)

ABSTRACT

In production systems, natural products have been outstanding in replacing synthetic products, among them, we can highlight the vegetable oils rich in polyunsaturated fatty acids (PUFA) that are popularly known for their beneficial effects. Thus, the objective of this study was to evaluate the effects of pequi (Caryocar brasiliense) and sunflower (Helianthus annus) oil supplementation on nursery physiological parameters. We used 180 piglets housed in a commercial farm, distributed in 3 groups (n=60) according to food supplementation: pequi oil, sunflower oil and negative control. Blood samples and animal weight were obtained at intervals of four nursery phase rations to evaluate performance, inflammatory response, lipid profile and oxidative status. The physiological parameters measured showed that the oils positively interfered in the systemic inflammatory response through total leukocytes and neutrophil / lymphocyte ratio (P<0.05), in the oxidant-antioxidant balance by measuring nitric oxide and malondialdehyde (P<0.05). and lipid metabolism with the assessment of total cholesterol and triglycerides (P<0.05). On the other hand, these vegetable oils interfered with weight gain and feed intake (P<0.05). Thus, it can be concluded that supplementation with pequi and sunflower oils improves animal health, but has a negative impact on the piglet's performance in the nursery phase.(AU)

ABSTRACT

Staphylococcus aureus is a pathogen associated with intrahospital infections commonly found in the nasal cavities and the hands of health personnel, as well as, on environmental surfaces; which become potential reservoirs and transmission vehicles of infections. In this study the frequency and susceptibility to penicillin and methicillin of environmental isolates of S. aureus in a hospital to Cuenca were analyzed. 50 samples (30 of two operating room and 20 of the intensive care room) were collected. S. aureus was identified by phenotypic tests and molecular detection of the nuc gene. The susceptibility to methicillin and penicillin was determined by agar disc diffusion method and the blaZ and mecA genes by polymerase chain reaction. The frequency of S. aureus was 6% (3/50 strains). Resistance to penicillin and methicillin was 66.6% (2/3 strains). The blaZ and mecA genes were detected in the two strains resistant to penicillin and methicillin. The low frequency of S. aureus may be related to the environments analyzed; because the surfaces sampled are areas where emphasis is placed on the application of hygiene and disinfection protocols for ensure adequate decontamination

ABSTRACT

Three medicinal plants with recognized anti-inflammatory potential, identified as “erva de São João“ (Ageratum conyzoides), “Tanchagem“ (Plantago major) and “ Bardana“ (Arctium lappa L.) were obtained from a medicinal herbs company located in Teófilo Otoni city (Minas Gerais State, Brazil). The dry plant material obtained in packages was submitted to the chemical procedures to prepare the crude extracts by maceration according to the Brazilian pharmacopoeia legislation. After extraction, the samples were subjected to 1H NMR, TLC and Capillary Electrophoresis analysis by co-injection of authentic patterns of phenolic acids and flavonoids to identify the major compounds and classes of secondary metabolites present in each material and then their chemical and biological potential was assessed by DPPH free radical inhibition assay and antimicrobial against E. coli. The results obtained allow us to conclude that the phytopreparation was effective in the extraction of compounds with antioxidant potential and the three species presented a high concentration of flavonoids and other phenolics that is compatible with the chemosystematic data. The screening obtained by 1H NMR spectroscopy, TLC and Capillary Electrophoresis with ultraviolet detection analysis provided us a qualitative profile of the phytochemicals present in each material. None of the extracts were active against Escherichia coli by antibacterial disk diffusion assay at concentration of 1 mg/ml.

ABSTRACT

Environmental enrichment is an important tool within production systems to promote welfare and animal health. The aim of this study was to evaluate the effect of enrichment objects on stress of piglets in nursery phase. 32 piglets housed in experimental farm were distributed in 4 groups (n= 8): rope, chain, pet bottle and negative control. Blood samples were collected at the beginning and end of the experiment to count circulating leukocytes and determine non-enzymatic antioxidants, nitric oxide, malondialdehyde, and saliva to evaluate cortisol. At the same time, an etogram and evaluation of mean daily gain and feed conversion were applied. The evaluated parameters on first day of the experiment did not vary between groups (P> 0.05). On the last day, neutrophil and neutrophil /lymphocyte ratios were higher in chain group piglets, as were salivary cortisol values (P< 0.05). Uric acid was higher in chain group and MDA in bottle group (P< 0.05). Environmental enrichment stimulated positive behaviors in piglets, where the rope stood out as the most attractive. On the other hand, chain had a negative effect on animal physiology, generating stress, as well as the bottle that induced lipid peroxidation and a lower weight gain in piglets.(AU)

ABSTRACT

Diarrhoeagenic Escherichia coli (DEC) is a leading cause of infectious diarrhoea worldwide. In recent years, Escherichia albertii has also been implicated as a cause of human enteric diseases. This study describes the occurrence of E. coli pathotypes and serotypes associated with enteric illness and haemolytic uremic syndrome (HUS) isolated in Brazil from 2011 to 2016. Pathotypes isolated included enteropathogenic E. coli (EPEC), enteroaggregative E. coli (EAEC), enterotoxigenic E. coli (ETEC), enteroinvasive E. coli (EIEC) and Shiga toxinproducing E. coli (STEC). PCR of stool enrichments for DEC pathotypes was employed, and E. albertii was also sought. O:H serotyping was performed on all DEC isolates. A total of 683 DEC and 10 E. albertii strains were isolated from 5047 clinical samples. The frequencies of DEC pathotypes were 52.6% (359/683) for EPEC, 32.5% for EAEC, 6.3% for ETEC, 4.4% for EIEC and 4.2% for STEC. DEC strains occurred in patients from 3 months to 96 years old, but EPEC, EAEC and STEC were most prevalent among children. Both typical and atypical isolates of EPEC and EAEC were recovered and presented great serotype heterogeneity. HUS cases were only associated with STEC serotype O157:H7. Two E. albertii isolates belonged to serogroup O113 and one had the stx2f gene. The higher prevalence of atypical EPEC in relation to EAEC in community-acquired diarrhoea in Brazil suggests a shift in the trend of DEC pathotypes circulation as previously EAEC predominated. This is the first report of E. albertii isolation from active surveillance. These results highlight the need of continuing DEC and E. albertii surveillance, as a mean to detect changes in the pattern of pathotypes and serotypes circulation and provide useful information for intervention and control strategies.

ABSTRACT

Pavonia alnifolia A.St.-Hil.

ABSTRACT Gastro protective activity of the Pavonia alnifolia A.St.-Hil. extract. In Brazil, the Malvaceae family is represented by at about 200 species. Some of those species are known as gastro protective ones. The Pavonia alnifolia A.St.-Hil (Malvaceae) was selected after a chemosystematic approach. The gastro preventive activity of the ethanol extract of stems Pavonia alnifolia was evaluated through the use of the Ethanol:chlroridric acid model on mice. The quantification of the total flavonoids (aluminum chloride method) and total polyphenols (Folin-Ciocalteu method) was also performed since the relation of those components with gastric protection has been previously highlighted. The tests showed a significant reduction of the ulcer formation in the mice treated with the plant extract (10, 100 and 300 mg/kg). This effect may be related to the presence of polyphenols whose content was found to be 74.3 ± 7.5 µg/mg of vegetal material and 82.7 ± 7.1 µg/mg of crude extract and flavonoids, which in turn showed a content of 17.1 ± 1.4 µg/mg dry extract

ABSTRACT

Sex hormones modulate the action of both cytokines and the renin-angiotensin system. However, the effects of angiotensin I-converting enzyme (ACE) on the proinflammatory and anti-inflammatory cytokine levels in male and female spontaneously hypertensive rats (SHR) are unclear. We determined the relationship between ACE activity, cytokine levels and sex differences in SHR. Female (F) and male (M) SHR were divided into 4 experimental groups each (n = 7): sham + vehicle (SV), sham + enalapril (10 mg/kg body weight by gavage), castrated + vehicle, and castrated + enalapril. Treatment began 21 days after castration and continued for 30 days. Serum cytokine levels (ELISA) and ACE activity (fluorimetry) were measured. Male rats exhibited a higher serum ACE activity than female rats. Castration reduced serum ACE in males but did not affect it in females. Enalapril reduced serum ACE in all groups. IL-10 (FSV = 16.4 ± 1.1 pg/mL; MSV = 12.8 ± 1.2 pg/mL), TNF-α (FSV = 16.6 ± 1.2 pg/mL; MSV = 12.8 ± 1 pg/mL) and IL-6 (FSV = 10.3 ± 0.2 pg/mL; MSV = 7.2 ± 0.2 pg/mL) levels were higher in females than in males. Ovariectomy reduced all cytokine levels and orchiectomy reduced IL-6 but increased IL-10 concentrations in males. Castration eliminated the differences in all inflammatory cytokine levels (IL-6 and TNF-α) between males and females. Enalapril increased IL-10 in all groups and reduced IL-6 in SV rats. In conclusion, serum ACE inhibition by enalapril eliminated the sexual dimorphisms of cytokine levels in SV animals, which suggests that enalapril exerts systemic anti-inflammatory and anti-hypertensive effects.

ABSTRACT

There is little information about in vivo toxic effects of the ethanolic extract of leaves of Carica papaya L. (ECP). Therefore, in this study ECP was characterized chemically by HPLC-RP and the antimutagenic and cytotoxic activities of an aqueous solution of ECP (CA) were assessed by the micronucleus (MN) bioassay. The extract consisted mainly of polar substances, one of which was rutin. The MN test was performed on groups of Wistar rats, as follows: negative control (NC) - vehicle; positive control (CP40) - cyclophosphamide (40 mg/kg, ip), 24h; extract-treated (CA) - ECP (500 mg/kg, po), 24h; extract + cyclophosphamide-treated (CACP40) - ECP (500 mg/kg, po), 48, 36 and 24 h, plus cyclophosphamide (40 mg/kg, ip), 24h. The MN index was 3.2 ± 1.79 and 1.6 ± 0.5, for NC and CA, and the PCE-to-NCE ratio was 1.38 ± 0.52 and 1.13 ± 0.28, respectively, indicating low cytotoxicity of CA. CP40 showed a high MN index of 20 ± 4.9, but CACP40 only 3.0 ± 1.6, the same as NC, indicating an antimutagenic effect. The study suggests that ECP has low toxicity and possesses an antimutagenic protective effect in which rutin may be involved.

ABSTRACT

The aim of the present study was to compare healing obtained with biomembranes with the natural healing process (sham) using biochemical and immunohistological assays. C57BL/6 mice were divided into 4 groups of 15 mice each and received different subcutaneous implants: natural latex biomembrane (NLB), denatured latex (DL), expanded polytetrafluorethylene (ePTFE), or sham. On the 2nd, 7th, and 14th days post-treatment, 5 mice per group were sacrificed and biopsied for the following measurements: oxidative stress based on malondialdehyde (MDA), myeloperoxidase (MPO) and hydrogen peroxide by the method of ferrous oxidation-xylenol orange (FOX), as well as glutathione and total proteins; histological evaluation to enumerate inflammatory cells, fibroblasts, blood vessels, and collagen, and immunohistochemical staining for inducible nitric oxide synthase, interleukin-1β, vascular endothelial growth factor (VEGF), and transforming growth factor-β1 (TGF-β1). On day 2 post-treatment, NLB stimulated a dense inflammatory infiltrate mainly consisting of polymorphonuclear cells, as indicated by increased MPO (P < 0.05), but oxidative stress due to MDA was not observed until the 7th day (P < 0.05). The number of blood vessels was greater in NLB (P < 0.05) and DL (P < 0.05) mice compared to sham animals on day 14. NLB induced fibroplasia by day 14 (P < 0.05) with low expression of TGF-β1 and collagenesis. Thus, NLB significantly induced the inflammatory phase of healing mediated by oxidative stress, which appeared to influence the subsequent phases such as angiogenesis (with low expression of VEGF) and fibroplasia (independent of TGF-β1) without influencing collagenesis.

ABSTRACT

A espécie vegetal Alpinia zerumbet (Pers.) B.L.Burtt & R.M. Sm.

Alpinia zerumbet (Pers.) B.L. Burtt & R.M.Sm. is traditionally employed to treat several diseases such as hypertension. The aim of this study was to evaluate the chemical composition, the anti-hypertensive activity and the capacity to reduce cardiac hypertrophy of the essential oil of A. zerumbet leaves (EOAZ) in rats. EOAZ was obtained through hydrodistillation in Clevenger apparatus and its chemical composition was analyzed by gas chromatography-mass spectrometry (GC-MS). Several constituents (14) were identified, terpen-4-ol (37.45 percent) being the major component, followed by caryophyllene oxide (7.56 percent), trans-sabinene hydrate (6.61 percent) and 1,8-cineol (4.02 percent). The cardiovascular effect was investigated after chronic treatment with spontaneously hypertensive rats (SHR) and their respective controls, Wistar-Kyoto rats (WKY). The treated group showed a lower mean arterial pressure (MAP) (SHRP: 160 ± 7 mm Hg; p<0.01) than the untreated group (SHR: 180 ± 5 mm Hg). The ratio of left ventricle-to-body weight (LV/BW) for SHRP was lower (2.504 ± 0.03 mg g-1; p<0.01) than that for SHR (2.162 ± 0.01 mg g-1), confirming the cardiac hypertrophy (CH) reduction. There were significant differences in MAP and CH between SHRP animals and control rats (WKY: 116 ± 2 mm Hg and WKYP: 119 ± 4 mm Hg; p<0.05. WKY: 2.152 ± 0.04 mg g-1 and WKYP: 2.168 ± 0.04 mg g-1; p<0.01), indicating that these values were not normalized. Those data showed that the chronic treatment with EOAZ reduces MAP and CH in SHR probably due to the presence of the compounds terpinen-4-ol and 1,8-cineol. Studies with higher doses or longer treatment periods are necessary to evaluate whether EOAZ can reduce the analyzed parameters (MAP and CH) to normal values.

ABSTRACT

,Curitiba-PR). Entre as vantagens observadas, vale salientar a menor morbidade, menor tempo cirúrgico, maior previsibilidade e facilidade na realização do procedimento cirúrgico.

Oral rehabilitation in totally edentulous patients by means of implantology and prosthodontics has been making progress with the developments in surgical, prosthetic and imaging techniques. With the advances in the technology employed in imagining equipment, surgery for the placement of dental implants can be performed with the aid of computed technology converting the images through specific software, making it possible to obtain previously planned surgical prototypes and guides, thereby making the surgical procedures for prosthetic rehabilitation more foreseeable and less traumatic. This paper makes a review of the literature, addressing current aspects of guided surgery for dental implants, and reports a clinical case of rehabilitation of total tooth loss in the maxilla, using the Neoguide System (NEODENT, Curitiba-PR). Among the advantages observed, the following are particularly worthy of mention: lower morbidity, shorter duration of surgery, greater predictability and ease in performing the surgical procedure.

ABSTRACT

Obstructive apnea (OA) can exert significant effects on renal sympathetic nerve activity (RSNA) and hemodynamic parameters. The present study focuses on the modulatory actions of RSNA on OA-induced sodium and water retention. The experiments were performed in renal-denervated rats (D; N = 9), which were compared to sham (S; N = 9) rats. Mean arterial pressure (MAP) and heart rate (HR) were assessed via an intrafemoral catheter. A catheter was inserted into the bladder for urinary measurements. OA episodes were induced via occlusion of the catheter inserted into the trachea. After an equilibration period, OA was induced for 20 s every 2 min and the changes in urine, MAP, HR and RSNA were recorded. Renal denervation did not alter resting MAP (S: 113 ± 4 vs D: 115 ± 4 mmHg) or HR (S: 340 ± 12 vs D: 368 ± 11 bpm). An OA episode resulted in decreased HR and MAP in both groups, but D rats showed exacerbated hypotension and attenuated bradycardia (S: -12 ± 1 mmHg and -16 ± 2 bpm vs D: -16 ± 1 mmHg and 9 ± 2 bpm; P < 0.01). The basal urinary parameters did not change during or after OA in S rats. However, D rats showed significant increases both during and after OA. Renal sympathetic nerve activity in S rats increased (34 ± 9 percent) during apnea episodes. These results indicate that renal denervation induces elevations of sodium content and urine volume and alters bradycardia and hypotension patterns during total OA in unconscious rats.

ABSTRACT

Garlic (Allium sativum) has several beneficial effects on the cardiovascular system, such as reductions of the levels of total cholesterol, LDL-cholesterol and blood pressure, besides acting as an antioxidant. The aim of this study was to assess the effects of administering Allium sativum on changes in the cardiovascular hemodynamics and macroscopic structure that occur in the hearts of animals with experimentally induced cardiac infarction. Male Wistar rats were treated with homogenized garlic at a dose of 125mg/kg b.w./day for 21 days, given orally for one week before and two weeks after the procedure to induce myocardial infarction. The control group was subjected to a fictitious surgery (SHAM). The animals were divided into control and infarcted groups, treated (SHAMT, INFT) or untreated (SHAM, INF) with garlic. There were reductions in right ventricular hypertrophy (INF=0.75±0.05 vs. INFT=0.61±0.03 mg.kg-1; p<0.01) and infarcted area (INF=29.7±4.8 % vs. INFT=20.4±1.4 %; p<0.05) and regularization of the levels of systolic (SAP; INF=100±8 vs. INFT=127±7 mm Hg; p<0.05) and mean arterial pressure (MAP; INF=94±4 vs. INFT=110±6 mm Hg; p<0.01) in the INFT animals, compared to the INF group. Fewer animals died after the cardiac infarction procedure in the group INFT than in INF (20%, n=2; 45.5%, n=5; respectively). These findings suggest that garlic can have an important role in the prevention and control of cardiovascular abnormalities, since there was a reduction in the number of post-infarction deaths and an improvement of the cardiovascular profile in the INFT animals.

ABSTRACT

We describe the clinical and molecular characteristics of two unrelated Brazilian families with an association of the Sicilian form of (deltaß)º-thalassemia with hemoglobin S and ß-thalassemia. Direct sequencing of the ß-globin gene showed only the hemoglobin S mutation in patient 1 and the ß-thalassemia IVS1-110 in patient 2. The other allele was deleted in both patients and PCR of DNA samples of the breakpoint region of both patients showed a band of approximately 1,150 bp, expected to be observed in the DNA of carriers of Sicilian (deltaß)º-thalassemia. The nucleotide sequence of this fragment confirmed the Sicilian deletion. There are few reports concerning the Hb S/(deltaß)º-thalassemia association and patient 2 is the first reported case of Sicilian type of (deltaß)º-thalassemia in association with ß-thalassemia documented at the molecular level

ABSTRACT

The effect of electrolytic lesion of the median raphe nucleus was measured on behavioral and physiological parameters related to stress 24 h after the lesion. In of the elevated plus-maze the lesion decreased the percentage of open arm entries and tended to shorten the time spent on the open arms indicating as increase in anxiety. In contrast, the lesion markedly increased the time spent in the bright (aversive) compartment of the light-dark box and decrease in attempts to cross from the dark toward the bright compartment, an anxiolyic effect. With the exception of plasma prolactin level, which was lowered by the lesion, the physiological measures used in the present study indicate that the lesioned animals are under stress. Thus, death rate and weight loss after the surgery were higher in lesioned than in control animals. In addition, lesioned animals showed higher plasma corticoster- one levels, a high incidence of gastric ulcers in the fundus and a depressed immune response to the mitogen concavaline A. These results highlight the importance of the median raphe nucleus in the regulation of stress and anxiety. They also show that behavioral and physiological measures of stress may be dissociated.

ABSTRACT
